天目铁木OrTIP1同源基因在拟南芥中的异源转化和功能初步研究
摘要:天目铁木(Ostrya rehderiana Chun)属桦木科(Betulaceae)铁木属(Ostrya)落叶乔木,是浙江省特有的极小种群野生植物,野生植株仅存5株,5株植株中仅1株能够正常结种、种子数量较多,其余4株个体结种数量极少甚至不结种,存在育性高低差异。分布地域狭窄、人为干扰和气候变化应是天目铁木种群规模急剧减小的直接原因;极小的种群规模导致其近亲交配更为频繁,遗传多样性降低,进而导致其生殖生长发育存在潜在的缺陷;加之天目铁木所处群落中其他种群的资源竞争,幼苗自然更新存在很大困难,种群规模难以恢复。经过前期对天目铁木育性的研究,我们对不同育性野生天目铁木个体的雌花序进行了转录组表达差异分析,发现并克隆了一个与天目铁木育性相关的基因OrTIP1,通过异源转化获得了转基因拟南芥植株。本研究拟利用获取的转基因拟南芥,结合TIP1同源基因的相关研究报道,探究天目铁木该同源基因的潜在功能和与育性之间的相关性,为天目铁木的致濒机制提供理论依据。

Preliminary study of heterologous transformation and function of Ostrya rehderiana OrTIP1 orthogs in Arabidopsis
Abstract: Ostrya rehderiana Chun is a unique wild plant in Zhejiang Province. There are only 5 wild plants, only 1 of 5 plant can breed normally and have a large number of seeds. The remaining 4 individuals have very few or no seeds, shows a difference in fertility. The narrow distribution area, anthropogenic disturbance and climate change are the direct causes of the decreasing population size; Extremely small population size causes more frequent inbreeding and reduced genetic diversity, and leads to potential defects in reproductive growth and development; With the competition of other populations of species, itrsquo;s difficult to recover the population size. With the previous studies of fertilities of Ostrya rehderiana Chun, we performed transcriptome expression analysis of female inflorescences from different wild individuals, found a OrTIP1 gene which related to its fertility and cloned it. The transgenic Arabidopsis plants by heterologous transformation were obtained. This study intends to use these transgenic Arabidopsis, combined with the related study of TIP1 homologous genes, to explore its potential function and relationship with fertility, to provide a theoretical basis for the endangered mechanism of Ostrya rehderiana Chun.
