Purpose and background of the research
- Purposes
1) To study the inhibition of Red Golgin Berry (RGB) on MAO A activity of LNCaP and C4-2B cell lines.
2) To study the effect of RGB extract on cell toxicity of LNCaP and C4-2B cell lines using Cell Number Assay.
3) To study the effect of RGB on mitochondrion function using Seahorse Assay.

- Significance
In the United States, the incidence of prostate cancer is on the increase and has become the second malignant tumor in men. Based on the research of Dr. Jean C. Shihrsquo;s group, monoamine oxidase (MAO A) is a mitochondria-bound enzyme that is highly expressed human prostate cancer and mediates prostate tumorigenesis and metastasis. And MAO A inhibitors (MAOAIs) were found to reduce the proliferation, migration and invasion of prostate cancer cells LNCaP, PC-3 and C4-2B. However, MAOAIs still have some side effects such as vision disturbance and blood pressure change. At present, the search for new drugs for prostate diseases from natural products has attracted the attention of scholars. Red Golgin Berry (RGB), the fruit of Lycium barbarum L., a well-known Chinese herbal medicine as well as valuable nourishing tonic, has been used as anti-inflammation and anti-aging agent for thousands of years. Modern pharmacological experiments have reported that the extract of RGB can inhibit the growth of prostate cancer cells. This study is aimed to find out if RGB has MAOAI activity using androgen-dependent LNCaP cells and androgen-independent C4-2B cells. We will test the RGB extract to study their effects on the cell toxicity, MAOAI activity and mitochondrion function of these two cell lines.

- Current achievements and prospects
Red Golgin Berry (RGB) is a red-orange berry of a Chinese herb Lycium barbarum (LB), commonly known as wolfberry. They are a popular functional food throughout the world with a large variety of beneficial effects such as immunomodulation, anti-aging, increasing male fertility and reducing blood sugar. Many studies have confirmed that RGB have many biological and pharmacological activities in treating various diseases, such as cancer, hyperlipidemia, hepatitis, and diabetes. For example, L.Gan et.al found that the polysaccharide in LB (LBP) could inhibit the growth of human leukemia HL-60 cells in vitro. JL Liu et. al found that LBP could dose-dependently decrease tumor weight in S180-bearing mice in vivo. Therefore, RGB has great potential for further development in pharmaceutical areas. However, the effect of RGB on prostate cancer cells have not been investigated clearly, which is one of the most common male malignant tumors in Western countries. Previous studies reported that RGB can induce spoptosis in many different types of cancer cells. In this study, RGB extract will be used to examine its influences on prostate cancer cell lines LNCaP and C4-2B. Besides, most of these researches were investigated in vitro or in mouse model, we expect future studies to elucidate the structure of active component and the relationship between bioactivities and chemical structure, which would allow a better understanding of this Chinese herb.
